It's just misfire - fuel doesn't ignite consistently at first when everything is cold. Fuel buildup can cause it to ignite back into the intake manifold. In your case it's pretty harmless, it should last only a short while and backfires are minimal, just let it idle it out. In other cases it can be a problem - first thing that comes to mind is the early 80's porsche 911 SC with the CIS fuel injection systems. Intake backfire would split open the airbox!

I strongly enriched several start tables and got first-try starts on e85 at 50F. This is with Propel, which has a high ethanol content. A gallon or two of 91 will also go a long, long way towards easing starts. If you go from a near-empty tank of 91 and then fill it with ethanol, you'll start just great because of that gallon of gas sloshing around. Run it low again and fill up with e85, however, and without that extra gas, the hard starts begin.

With true E85 I had problems starting below 50 degrees with a unaltered OFT Stage 2 E85 tune.

Someone will have to define "hard start". To me that means it doesn't start on the first crank. To someone else that might mean it starts after 5 or 10.

Mine's a SmartKey so I go by if it starts on the first button press or if I have to shut it off and try again; havent had to do that since making the IPW changes
