I'd typically run the tank down as close to empty before switching over to an E85 tune. Fueling up close to 11 gallons, of E85. Id start the car up after topping the tank and watch the fuel trims as I putt along or just let it idle. Once sufficiently "maxed out" the fuel trims are, stop the car and flash the E85 tune. The rest is just like any old reflash. You might still not be up to E85 but should be pretty close by the second tank of e 85.
The lower you run the tank down before switching, the less likely you will run in to unstable or high fuel trims.
