Ecutek like romraider or ecuflash is just a system to alter the parameters (tables) so you can change the input parameters to these calculations.

Ecutek does add some extra code to ecu, but unless your using speed density fueling, your likely using the standard fueling as calculated by the ecu

This would not be a simple calculation as the ecu looks ant many inputs including MAF , and many tables in the ecu to arrive at the amount of fuel its going to inject via the two fueling systems.

If you wanted your car to run on E85 all you need to do is alter the port and direct injector "scaling/pressure multipliers, so the ecu would inject 30% more fuel, the calculation the ecu would perform is the same but you "fooled" the ecu into injecting 30% more fuel as you want to run E85. Compare the OFT petrol and E85 tunes, thats all that done fueling wise. Ecutek its basicly same thing
If we tune E85 the ecutek is just a tool but i guess flexfuel feature in ecutek has internal calculation for E85.
