The thing to remember is that the E85 tune is more aggressive.
What this means is that if you accidentally run E85 on the gasoline tune you won't hurt anything, you just won't make power. The car will feel sluggish.
If you do the reverse and run gasoline on the E85 tune there is a chance of damaging something, although even then the ecu should pull timing to protect the motor.
If you are switching from gas to corn and are seriously paranoid, you could fill up with E85 and drive for a few miles with the gas tune then flash to the E85 tune. From what I've read you should know when the lines are clear of gasoline.
All of this is based on reading: I don't have E85 readily available so no personal experience with it.
