This might be a dumb question but will my custom tune be lost if I disconnect the battery?

No

its permanent till ecu is re flashed

disconnecting battery will just reset learned parameters to default and clear CEL codes

see flashing -Reset - Learning link below

Whats xdf? This tune number is the eu2 tune for manual tranny. It took me about 15 mins to flash mine last year.
