There are stronger gear sets available for our transmission, why not just do that. Or CD009.

Lol my 4th gear and 5th already feel like garbage. The gear sets I have seen cost about 7k that's almost half of an LS swap at that point.

Close to 280 to the wheels on E85, but my 4th and 5th gear are grinding already. Bought a hydraulic TOB to see if that helps.

280 what, nm, lb/ft

This is what has stopped me from boosting mine, the Aisin is only rated to 250nm.

If its in lb/ft no wonder your eating 4th, thats 100nm over rated max.

How would a transverse transmission work with an inline engine? Is that possible?

Mazworx literally has a kit just for this that included a new CD009. Or you could just use another twin transmission for a 10th the cost.
